GENETIC VARIABILITY OF NON POTATO POTATO VIRUS Y ISOLATES IN ITALY

Abstract:
Serious outbreaks of Potato virus Y (PVY) on tomato crops were observed during the summers of 2002 and 2003 in southern Italy, with symptoms of mosaic and leaf distortion, stunting, flower necrosis and irregular fruit ripening, and a disease incidence ranging from 85 to 100% in some areas. Preliminary biological and serological investigations allowed us to establish that observed symptoms were associated with PVY infection. Analyses performed on wild plants in the areas of interest showed that Solanum nigrum is the most important host species for PVY in the field. Tomato and S. nigrum samples, representative of Basilicata, Campania, Calabria, Puglia and Lazio regions, were collected from different fields for the molecular characterisation of PVY isolates. RT-PCR was performed on viral RNA extracted from purified virion, using two couples of primers specifically designed on the coat protein of non potato PVY isolates. PCR products were then cloned and sequenced. Results of pair-wise similarity analysis reveals a closer relationship with the PVY isolates of the non potato group, with percentage values ranging from 92 to 96% at nucleotide level. This result is confirmed by phylogenetic analysis performed on the coat protein gene.
