CHARACTERISATION OF THE POTATO VIRUS Y ISOLATE PVY-LF02 INDUCING NECROSIS IN TOMATO

Abstract:
A serious viral disease of leaf necrosis, necrotic mottling along leaflet veins, rachis distortion and irregular fruit ripening was observed in hydroponic production of different tomato cultivars in the Calabria region of southern Italy. A virus with filamentous particles, about 750 nm long, was observed by electron microscopy in leaflet dips from naturally infected tomato plants from 15 different plots. This virus was mechanically transmitted from symptomatic plants to herbaceous indicators. Preliminary biological and serological investigations allowed to establish that observed symptoms were always associated with Potato virus Y (PVY) infection. In particular, symptomatic tomato samples were analysed by ELISA using monoclonal antibodies for discrimination between PVYN, PVYO and PVYC subgroups. RT-PCR was performed on viral RNA extracted from purified virions using primers specifically designed to the coat protein of PVYNP (non potato) isolates. PCR products were then cloned and sequenced. The necrotic syndrome was reproduced in tomato seedlings inoculated with PVY-LF02 isolate. Biological characterisation showed that the isolate induces vein banding and mottling symptoms in Nicotiana tabacum ‘Xanthi’ and ‘White Burley’ and no symptoms on Chenopodium amaranticolor and C. quinoa. Serological assays revealed that the isolate PVY-LF02 reacts with PVYC-specific Mab but pair-wise analysis does not allow association with any of the PVY groups established on a molecular basis. Our results indicate that PVY-LF02 is a PVY isolate in spite of its peculiar biological and molecular properties.
