INVESTIGATION OF GENETIC DIVERSITY AMONG EUROPEAN HAZELNUT (CORYLUS AVELLANA) CULTIVARS USING SSR MARKERS

Abstract:
We used 10 pairs of simple sequence repeat (SSR) primers to investigate genetic diversity in 272 cultivars of European hazelnut (Corylus avellana) representing a wide range of geographical regions. Of the 272, 80 are suspected duplicate accessions, as they are morphologically identical, but were imported from different collections under different names. Preliminary PCR results showed that the SSR loci we used are highly polymorphic. ABI Genescan® and Genotyper® software were used to identify the precise allele size generated from each PCR reaction based on an internal lane standard. PowerMarker software was used to generate a genetic similarity matrix based on possible pair-wise combinations of accessions using the “proportion of shared alleles”. UPGMA cluster analysis was used to construct and draw a tree from the genetic similarity matrix using PowerMarker and Tree View software. The results are discussed.
