CHARACTERIZATION OF ARTICHOKE OPEN POLLINATED AND HYBRID CULTIVARS BY PROTEIN ELECTROPHORESIS

Abstract:
Little research-based information is available on artichoke (Cynara cardunculus L. subsp. scolymus Hayek) genetic variability achieved by biochemical and molecular markers. Studies on artichoke cultivars characterization by achene proteins analysis are unknown. The pattern of achene proteins, assessed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE), can be utilized as marker in open pollinated (OP) cultivars characterization as well as in the identification and genetic purity checking of hybrids. In order to verify the usefulness of this technique in the artichoke characterization, ten OP and three hybrids were tested. Different patterns were identified on the basis of 18 polymorphic bands with medium to strong intensity but not correlated among them. Patterns showing at least one different band were considered different. In the OP cultivars 37 different patterns were identified. The number of patterns in each OP cultivar ranged from 4 to 12; in different OP cultivars, identical patterns with significantly different frequencies were found. Two hybrids were identified by only one pattern, whereas the third one was characterized by 2 patterns with frequency of 94% and 6%, respectively. The polypeptidic composition of achenes was able to reveal variability within and among analyzed cultivars. The technique under study can be also applied in genotypes hybrid identification and in estimation of their genetic purity. When artichoke cultivars cannot be distinguished by means of this technique other biochemical or molecular markers must be used.
