DEVELOPMENT OF MICROSATELLITE MARKERS FOR MARKER-ASSISTED BREEDING IN PEARS (PYRUS SPP.)

Abstract:
With the construction of a reference linkage map for apple (Malus spp.), consisting primarily of microsatellite markers, it is now possible to exploit this information to develop genetic maps for pear (Pyrus spp.) also, and thus develop markers for use in marker assisted breeding programs. A cross-species survey has shown that it is possible to amplify microsatellite repeats (SSRs) using Malus primers on Pyrus. We are currently implementing SSRs obtained through literature for Malus and Pyrus. In addition, we are developing new SSR markers from the analysis of genomic and cDNA sequences from Malus and Pyrus. Candidate SSRs have been identified with Tandem Repeats Finder, and then optimized for PCR and tested for polymorphism in both apple and pear. Markers are then multiplexed together for higher throughput screening and mapping.
