Abstract:
The gene for super sweet protein thaumatin II from Thaumatococcus danielli has been transferred to apple in attempts to improve taste and phytopathogen resistance.
Based on the coding sequence, cloned at Unilever, and pBI121, we constructed the binary vector for its expression in plants.
The successful transformation of the apple cultivar ‘Melba’ has been obtained by using the disarmed supervirulent Agrobacterium tumefaciens strain CBE21. The introduction of the thaumatin gene was confirmed by PCR analysis and its expression by Western blotting.
Leaves from transgenic lines demonstrated taste modification, detectable by an organoleptic test.
