Abstract:
The use of microsatellites for genotyping can occasionally be complicated by the preferential amplification of some alleles.
When PCR was performed on an apple mapping progeny using primer CH04f04 at the published annealing temperature, both parents appeared to amplify the same allele.
Some of the seedlings, however, amplified a different allele from that observed in the parents.
We have revealed the missing bands and have elucidated the true segregation for this locus by reducing the annealing temperature.
Markers showing similar problems are best avoided in paternity analyses and diversity studies.
