Abstract:
The use of phosphomannose isomerase (pmi) as selectable gene and mannose as selection agent was tested on leaf explants of the apple (Malus domestica) rootstock M26. Leaf explants could tolerate up to 2 g/l with or without agrobacterial infection.
In general, the shoot number per regenerated leaf was reduced with increasing concentration of mannose.
One transformed shoot was obtained carrying the pmi, gus and gai genes, confirmed by Southern blot analysis.
Mannose could be used as selection agent but the selection efficiency seems not as high as kanamycin.
However, more studies are needed to confirm this.
