Abstract:
Herbicides are widely used in nurseries and orchards.
Herbicide-resistant rootstocks for fruit production are a new way to enhance fruit crop safety and production.
The bar gene cloned in the RAS Bioengineering Center, has been used in our research for obtaining the phosphinotricine resistant apple clonal rootstock №545. Based on the development of transformation protocols, about 30 regenerants successfully proliferated on selective media supplemented by PPT were obtained. РРТ was inefficient as a selective agent because of a high frequency of escapes.
Only one transgenic line successfully proliferated on medium with 15 mg/L PPT (control plants died on 2-5 mg/L) and incorporation of the bar gene in its genome was confirmed by PCR analysis.
The transformation frequency was 1.3%. The transgenic plants were resistant to treatment by the Basta commercial herbicide in greenhouse and field conditions.
