Abstract:
The Ma resistance gene in Myrobalan plum (Prunus cerasifera L.) confers a full-spectrum and high-level resistance to root-knot nematodes (RKN). This diploid rootstock species is highly heterozygous, self-incompatible, and can be easily crossed with almond or peach.
Using a mapping population of approx. 300 intra and interspecific individuals derived from the resistant heterozygous accession P.2175, the Ma gene was located on the Prunus linkage group 7 between the markers SCAL19 and pchgms6 in a 2.7 cM interval.
For a map-based cloning approach, AFLP markers tightly linked to the Ma1 allele from P.2175 were obtained using a BSA strategy specifically targeting the interval between the previous markers.
Three of them were transformed into codominant SCAR or CAPS markers. Ma and the SCAFLP2 cosegregating marker were then finely mapped in a 0.8 cM interval between the nearest flanking markers SCAFLP3 and SCAFLP4 by extending the mapping population with c. a. 1000 offspring from P.2175. Then the construction of a P.2175 BAC library allowed the detection of a single BAC clone encompassing Ma.
