ISHS Acta Horticulturae 657: XIX International Symposium on Virus and Virus-like Diseases of Temperate Fruit Crops - Fruit Tree Diseases

EVALUATION OF THE SUSCEPTIBILITY OF SEEDLINGS FROM APRICOT CROSSES TO THE MARCUS STRAIN OF PLUM POX VIRUS

Abstract:
The potential risk of an epidemic spread of Plum pox virus (PPV) in the main Italian apricot-growing areas led to studies for the identification of natural sources of resistance to PPV.
Results of trials for evaluating the behaviour of some apricot seedlings, derived from crosses of cultivars considered to be tolerant, are reported. 250 accessions, provided by the Universities of Milan, Bologna and Pisa, were grafted onto myrobolan rootstocks, and graft-inoculated with isolate 0019-GR of the Marcus strain of PPV.
Plants, grown in insect-proof screen-house, were submitted weekly to visual inspections and tested twice by ELISA in one year period. Most of the accessions showed symptoms and were ELISA-positive, whereas 24 selections were symptomless but ELISA-positive. Fifteen accessions that were symptomless and ELISA-negative proved to be infected by PPV only when tested by RT-PCR.
