Abstract:
Fruit showing circular, sunken black necrotic lesions and circular ring symptoms from a single plum tree (Prunus domestica ‘Victoria’) from the Waikato region, North Island was submitted to Ministry of Agriculture and Forestry.
Flexuous filamentous particles were observed by electron microscopy, suggesting the presence of Plum pox virus (PPV). However, the fruit tested negative for PPV and potyviruses by ELISA, RT-PCR, and IC-RT-PCR. Subsequently, RT-PCR and IC-RT-PCR for Apple chlorotic leaf spot virus (ACLSV) was performed and a positive result was obtained, confirming the observation of filamentous particles.
The sunken necrotic lesions were consistent with infection by the bacterium Xanthomonas arboricola pv. pruni. Suspect samples collected within a 12 km radius of the initial site all tested negative for PPV.
