AGROINOCULATION AND PCR ANALYSIS COULD BE USED IN QUANTITATIVE ASSESSMENT OF SUSCEPTIBILITY TO VIRUS INFECTION

Abstract:
Due to lack of success in mechanical inoculation, and the low efficiency of particle bombardment inoculation, Agrobacterium tumefaciens cells carrying a partial tandem repeat construct of Strawberry vein banding virus (SVBV) were used to inoculate strawberry cultivars to assess their susceptibility to SVBV. An infection rate of 75-100% was achieved with agroinoculation of 23 different indicators and cultivars of strawberry (Fragaria spp.). Infected cultivars differed in symptom severity and had variable virus titers as quantified by PCR analyses. An ornamental strawberry (F. x ananassa ‘Temptation’) had the lowest titer while several commercial cultivars such as ‘Carlsbad’ had the highest titer of SVBV.
