MAPPING CANDIDATE GENES AND QTLS FOR AROMA CONTENT IN GRAPE

Abstract:
The importance of aroma component on wine quality and wine characterization makes genomics tools coupled with high resolution description of metabolic situation very useful to provide new insights into the varietal flavour of grapes. The availability of linkage maps also allows a wide range of studies, including identification and positional cloning of important genes and the understanding of genetic factors underlying quantitative traits (QTLs). A mapping approach was used to investigate the genetic control of principal compounds (terpenes) characterizing the aroma of the berries in an interspecific grape progeny. To obtain a suitable population for segregation analysis the intensely flavoured Vitis vinifera cv. Moscato bianco was crossed with an accession of Vitis riparia tolerant to important fungal diseases. Both free and glycosidically bound terpenes were detected by HRGC-MS and the marker-trait co-segregation analysis was performed by MapQTL 4.0 software using the maternal linkage map. Based on grape ESTs, candidate genes involved in the terpenoid biosynthesis in plants were tested for polymorphism and segregation. Putative QTLs for berry content of monoterpenes were identified.
