STUDY OF THE INFLUENCE OF MAJOR IONS AND BENZYLAMINOPURINE ON PROLIFERATION IN VITRO OF MICROSHOOTS OF APPLE, PLUM, TRIFOLIATA ORANGE AND BLACK CURRANT BY A SYSTEMATIC VARIANCE METHOD

Abstract:
In order to study the influence of macro-ions and benzyl aminopurine (BAP), they were supplied individually in high concentration while all other ions were not changed from a base level. These ion combinations were applied with or without BAP at the following concentrations to various fruit species: for apple rootstocks M-7 and M-9, 0,1-3mg/l; for MM-106 and Marubakaido, 0,1-4 mg/l; for plum Kantimirov­skaja, 0,1-1,5 mg/l; for Trifoliata orange, 0,1-1 mg/l and for Currant Zagadka, 0,1-2 mg/l. On medium with a high concentration of BAP, the most significant influence upon multiplication of the rootstocks M-7, MM-106 and Marubakaido was demonstrated by the ions NO3-, K+, NH4+; for M-9, the ions NO3-, NH4+, Ca+2; for plum,the ions NO3-, NH4+, K+, Ca+2; for Trifoliata orange , the ions K+, Ca+2 and for Currant Zagadka, the ions NO3-, NH4+, Ca+2 significantly increased proliferation. For shoot elongation of rootstocks of apple M-7 and Trifoliata orange the ions K+, NO3-; for rootstock of apple Marubakaido, the ions NO3-, Ca+2; for rootstocks of apple M-9, MM-106 and plum Kantimirovskaya the ions NH4+, NO3-, K+, Ca+2 were more effecient. For shoot growth, higher level of BAP were very important; for plum Kantimirovskaya 0,5 mg/l; for rootstocks of apple M-7, M-9 and Trifoliata orange 1 mg/l; for rootstocks of apple MM-106 and Marubakaido 2 mg/l. At these optimum levels of BAP, the ions NO3-, K+, NH4+ for rootstocks of apple M-9, MM-106 and Marubakaido; NO3-, Ca+2, and K+ for M-7 and plum Kantimirovskaya ; the ions NO3-, NH4+, K+, Ca+2 for Trifoliata orange; the ions K+, NO3- and NO3-, NH4+, Ca+2 for Currant Zagadka provided the greatest influence. With increasing concentration of BAP, shoot elongation increased. These concentrations of BAP stimulated development of more buds on explants with ions combinations NH4+, NO3-, K+ for apple rootstock Marubakaido, ions NO3-, K+, Ca+2 for M-9, MM-106 and plum Kantimirovskaya, ions NH4+, NO3- for rootstock M-7, ion K+ for Trifoliata orange and ions NO3-, NH4+, K+, Ca+2 for Currant Zagadka. Increasing the concentration of the ions SO4-2 and PO4-3 resulted in low rates of multiplication for these rootstocks.
