MICROPROPAGATION OF RHODODENDRON UWAENSE, AN ENDEMIC AND RED-DATA (ENDANGERED/RARE) SPECIES IN JAPAN WITH COMMERCIAL PRODUCTION POTENTIAL

Abstract:
Rhododendron uwaense is a new species to horticulturist. This species has been recently named and has a very limited distribution only to Tsushima town, Ehime Pref. in Japan. Blooms are pale purple to pink, medium sized (3-4 cm in diameter)in mid spring, and its fragrance often spreads across the valley. The population size is quite small, and it is considered a threatened species (IB-EN species) of Japanese flora. To introduce this species as a new ornamental plant, production of a large amount of nursery stock is needed. For this purpose, a micropropagation trial has been carried out to rapidly multiply this plant on a large scale. A two step tissue culture protocol has been developed for this species. Pre-culture was done using NAA (0.5 mg L-1) and TDZ/2ip (0.5 to 1.0 mg L-1) combined medium for inducing calli (Callus induction culture) for one to two months. Regeneration culture was done using 2iP medium without NAA (Shoot induction culture) using transplants from callus induction culture. Regenerated new shoots were obtained in 6 months.
