Abstract:
The highest percentages of explants with protocorm-like bodies (PLB) were observed on Vacin and Went (VW) basal medium supplemented with 0.1 NAA+0.5 BA (53.3%) for Earsakul, 0.5 NAA+0.5 BA (46.7%) for Omyapink and 0.5 NAA+0 BA (46.7%) for Semialba.
The highest percentage of explants with plantlets were similar for MS (Murashige and Skoog) and VW basal mediums.
The optimal culture temperature and sucrose concentration for PLB increase was 25C and 20g·L-1 respectively for all three cultivars.. However, plantlets regenerated from PLB that were cultured at 22C and 31C turned brown and had short shoots. Combinations of apple and tomato homogenates were more effective for plantlet growth than coconut water.
