EXPRESSION ANALYSIS OF GENES ENCODING AQUAPORINS DURING THE DEVELOPMENT OF PEACH FRUIT

Abstract:
To clarify the mechanisms of controlling water transport in peach fruit, the expression pattern of aquaporin genes (Pr-gTIP1, Pr-dTIP1, Pr-PIP1 and Pr-PIP2) were investigated. According to genome gel DNA blot analysis, each of those genes exist as a single copy in the peach genome. The amino acid sequence of Pr-gTIP1, deduced from the full sequence of the cDNA, was highly homologous to those of tonoplast intrinsic proteins (TIPs) isolated from other plants. RNA gel blot analysis revealed that Pr-PIP1, belonging to a PIP family, is constitutively expressed throughout development of fruits, while other aquaporins such as Pr-gTIP belonging to γTIP was mainly expressed at an early stage of fruit growth. The expression of Pr-gTIP1, Pr-dTIP1 and Pr-PIP2 decreased in response to drought stress, whereas that of Pr-PIP1 was not affected by the stress. These results suggest membrane permeability may be controlled by the down regulation of these aquaporins.
