DATE PALM (PŒNIX DACTYLIFERA) OFFSHOOT IDENTIFICATION BY PCR-ISSR MARKERS

Abstract:
Development of cultivar specific genetics markers is desirable for cultivar identification and protection. Intermicrosattelite amplification. ISSR (Inter Simple Sequence Repeat). PCR amplification of genomic DNA was carried out with primers which carry repeated dinucleotide constructs and some arbitrary primers. The amplified fragments are situated between microsatellite loci. The analysis had two objectives: to detect polymorphism within cultivars of date palm and to investigate the genetic stability of regenerated plantlets via somatic embryogenesis. The ISSR approach revealed highly reproducible polymorphism among the different varieties tested.
