Abstract:
Basal ends of micropropagated hazelnut shoots were dipped in 1000 ppm IBA (indole-3-butyric acid) solution for 5 or 10 sec, and were then cultured on plant growth regulator (PGR)-free Woody Plant Medium (WPM), or after dipping in IBA solution for 10 sec, microshoots were placed directly in peat plugs; all were then placed in culture vessels with 100% relative humidity but containing no plant nutrients or sugar.
Over 70% of shoots cultured on WPM rooted within two weeks, and 98% of shoots in the peat plugs rooted between 15 to 45 days.
Rooted microshoots were transplanted to pots, and then grown in an acclimatization room for 30 days.
After acclimatization, survival rates of plants placed in the greenhouse were 62% and 53% for microshoots treated with IBA for 5 and 10 sec and rooted on WPM, respectively.
The survival rate for plants cultured directly in jiffy peat plugs was 98%.
