ISHS Acta Horticulturae 610: V International Symposium on Kiwifruit

PRESENCE OF PROTEINS, CALLOSE AND PECTINS IN UNGERMINATED AND GERMINATED POLLEN OF ACTINIDIA DELICIOSA

Authors:   M. Oliveira, I. Abreu
Keywords:   kiwifruit, pollen grain, pollen tube, toluidine blue, aniline blue, alcian blue.
Abstract:
The principal components of pollen grain and tube wall were studied as well as the dynamic of these components during the pollen germination. In ungerminated and germinated pollen, proteins were detected using toluidine blue under light microscopy in the cytoplasm and intine, at the pore region and in the pollen coat. Pollen tube wall and callose plugs stained with aniline blue, but the pollen tube tip, which is the growth region of the pollen tube, did not stain. Finally, pectins were located, using alcian blue, in intine, at the pore region and in tube wall. In the growing pollen tube, pectins were also observed forming annular structures.
