Abstract:
Protein extracts from callus, young leaves, and berries of Vitis vinifera cv.
Nebbiolo were subjected to 2-D electrophoresis and the 2-D images were computer analyzed.
Different protocols of extraction and desalting were compared on callus extracts.
Extraction with an urea-based buffer and TCA/acetone precipitation gave the best results and were routinely used on all extracts.
The total number (between two and three hundred) of protein spots has higher in extracts from leaves, while the reproducibility of protein spots was higher in extracts from callus and lower in extracts from berries.
