Abstract:
In order to improve transient gene expression levels, suspension cells were bombarded with plasmid DNA at three concentrations (0.1, 0.5 or 1 µg DNA per shot). Two particule sizes (1.0 or 1.6 µm) also were shot onto cells deposited on medium containing 20 or 60 g.l-1 sucrose.
Results showed that high sucrose medium stimulated cell response while the larger particule size also induced more cells to GFP expression.
Experiments also indicated that higher level of expression could be obtained using half the control DNA amount
