Abstract:
Pear was regenerated through a callus stage and various factors were studied on shoot regeneration from leaf and petiole explants.
Placing the adaxial surface of leaf explants on regeneration medium improved shoot regeneration for ‘Burakovka’ but not for ‘Pamyat’ ‘Yakovleva’ or GP 217. In ‘Burakovka’, precultivation of explants in liquid regeneration medium was beneficial.
The effect of wounding explants by microparticles depended on genotype.
Leaf explants taken from the non-rooted, proliferation stage produced more regeneration in ‘Kosmicheskaya’ and ‘Burakovka’ than from rooted shoots but shoots of ‘Burakovka’ were vitrified.
Repeated cycles of culture and regeneration did not influence regeneration frequency.
