Abstract:
The effects of a 2,4-D pre-inoculation pulse on transformation efficiency of ‘Bartlett’ and ‘Abbé Fétel’ leaf explants were evaluated in order to develop a preliminary protocol for Agrobacterium-mediated transformation of pear.
The hormone conditioning treatment increased transformation efficiency, measured both as frequency of leaves with GUS activity and mean of GUS expressing foci/leaf based on late GUS histochemical assay results (recorded on day 35 and 135 after inoculation), and in contrast with earlier collected data (within day 20), the hormone conditioning treatment increased transformation efficiency, measured as both frequency of leaves with GUS activity and mean of GUS expressing foci/leaf.
