Abstract:
A molecular approach was applied to study pear/quince graft incompatibility using an innovative in vitro model.
The in vitro model includes a co-culture of pear calluses (cv.
Kaiser) with cell suspensions of pear (cv.
Kaiser) as controls (graft compatible) or with two rootstocks, the compatible clonal Pyrus communis rootstock Fox11 and the incompatible quince MC (EM-C or QC). The cultivars and rootstocks were physically separated by a solute permeable membrane.
The molecular approach applied was mRNA Differential-Display-Reverse-Transcriptase-PCR; this technique allows direct isolation and cloning of cDNA fragments corresponding to mRNAs differently expressed in the compatible or incompatible combinations.
The cDNA fragments, differentially expressed, were amplified and cloned.
The identification and the analysis of these cDNAs is in progress.
