Abstract:
The presence of susceptible and resistant European pear (Pyrus communis) cultivars to the ravages of fire blight suggests that it is possible to identify resistance gene sequences found in pear germplasm.
The present study reports the identification of pear RGA (resistance gene analogs) amplified from the fire-blight resistant pear genotypes Harrow Sweet, US 309 and Old Home using different heterologous primers designed in two conserved regions between different resistance genes.
PCR products were cloned and characterised by restriction enzyme digestion to identify different RGA classes.
At present, one clone in each of the most diverse RGA classes has been sequenced.
Gene bank database screening of pear RGA showed a certain degree of homology with known resistance gene sequences.
