FINGERPRINTING OF PRUNUS ROOTSTOCKS WITH MICROSATELLITES

Abstract:
Characterization of rootstocks of grafted trees is difficult or impossible using traditional methods. Consequently, molecular approaches should be implemented in rootstock identification programmes. In this work, twenty five genotypes useful as rootstocks for almond have been identified with microsatellite primers developed in peach. Eighteen primer pairs that resulted in correct and repeatable amplifications were sufficient to unequivocally identify all the genotypes studied. Those primer pairs produced a total of 95 alleles among the rootstocks studied. The results obtained show the usefulness of cross-species transportability of microsatellite sequences and the applications of microsatellites for fruit rootstock identification.
