Abstract:
This work aims to study the micropropagation of two pistachio species: Pistacia atlantica and P. terebinthus. Shoot tips and nodal segments taken from in vitro seedlings were cultured on MS and OM media and their dilution to the half (1/2 MS and 1/2 OM). Results showed little stimulation of shoot elongation and a reduction of shoot tip necrosis symptoms on OM medium for the two species.
With the objective of optimizing this medium, concentrations of macronutrients were reduced by a half (OM1/2) or multiplied by 1 (OM1), 3/2 (OM3/2) or 2 (OM2). This investigation proved the superiority of OM1 for P. atlantica and OM3/2 for P. terebinthus. These two media were supplemented with hormone mixtures containing 1, 2 or 4 ppm of BA. Growth kinetics relative to elongation has been studied in the two species according to the amount of BA applied.
The medium containing the high level of cytokinin produced many shoots from calluses developed at the basal part of the explant in the two species.
