Abstract:
In vitro grown shoots of 2 almond rootstock selections, the almond x peach rootstock GF677 and the peach cultivar 'Babygold', were cultured on the following multiplication media: MS (Murashige and Skoog, 1962) medium, MS medium with Fe-EDTA reduced to a 1/2 or a 1/4 or totally replaced by an equimolar amount of FeSO4 plus 1mM KHCO3. The calculation of multiplication rates was performed and chlorophyll content was determined.
Symptoms of Fe deficiency were more evident in presence of FeSO4 plus bicarbonates and clearly genotype dependent.
The most pronounced symptoms were noted in the peach cultivar 'Babygold', intermediate in GF677 and M51 and less evident in almond genotypes, suggesting a possible correlation between in vitro and field response.
These preliminary results support the possible use of tissue cultures for rapid screening of chlorosis tolerance in almond and Prunus spp.
