THE JAPANESE PEAR GENOME PROGRAM III. COPIA-LIKE RETROTRANSPOSONS IN PEAR

Abstract:
Fragments of copia-like retrotransposons were obtained using two degenerate primers corresponding to the conserved motifs of reverse transcriptase from pear, peach and apple. Fifty-one non-redundant sequences derived from Japanese pear ‘Housui’ (PhRT) were classified into 15 groups by 80% nucleotide identity. Phylogenetic survey revealed a high degree of heterogeneity among the groups. Southern blot analysis demonstrated that several types of retrotransposon-like sequences existed in the genomes of Pyrus species and polymorphisms were detected among Pyrus species as well as within the species. Copia-like retrotransposons were estimated to be present at about 1,000 copies and to account for 0.5~2.5% of pear genomes. Based on the sequence identities, the derivation of retrotransposons in pear, peach and apple was also discussed. The information on retrotransposons will contribute to the understanding of the genome structures and the principles of mutation in pear as well as other fruit tree species.
