AFLP MOLECULAR MARKERS OF 10 SPECIES OF PYRUS IN CHINA

Abstract:
DNA molecular markers of 10 species of Pyrus were conducted with AFLP (Amplified Fragment Length Polymorphism) technique. The results indicated that 355 DNA fragments were amplified by using 6 pairs, 9 combinations of primers, in which 281 fragments presented polymorphism and accounted for 79.2% of the total fragment numbers. On average, 39 DNA fragments were produced by a pair of primers and this presented the abundant polymorphism of Pyrus. Cluster analysis was carried out by using the amplified results of DNA. The analysis indicated that each species had different genetic distance. It was shown that AFLP technique can be used as the basis of the taxonomy and identification of species of Pyrus. The blood relationships among species of Pyrus were analysed at the DNA level.
