MOLECULAR BREEDING OF PEAR

Abstract:
Usage of hygromycin as selective agent was more effective in pear transformation than kanamycin in our experiments. Higher transformation frequency and lower escape formation were observed. The presence of intron in the coding region of a heterologous gene improved efficiency and stability of transgene expression in plant tissues. For fruit taste improvement the gene of supersweet protein thaumatinII from Thaumatococcus danielli was used in our research. Immunoblotting detectable level of thaumatinII protein in leaf tissues reached 0.25% from total plant protein. The organoleptical test of leaf tissue of greenhouse and field grown plants demonstrated taste modification. For plant protection against fungal and microbial attack, the plant defensins genes (PD) have been transferred to pear. Based on previously developed transformation methods, the PD gene from Raphanus sativus was transferred to pear variety Burakovka by using different agrobacterial vector systems. The bar gene cloned in the “Bioengineering Center” RAS has been used in our research for obtaining phosphinotricine (PPT)-resistant pear clonal rootstocks. Based on development transformation protocols, 17 transgenic PPT-resistant clones were obtained. Some of them were resistant to herbicide Basta treatment equivalent for 24 1/ha. All transgenic lines are tested in field plots since 2000 year.
