IDENTIFICATION OF SOME TURKISH OLIVE CULTIVARS BY USING RAPD-PCR TECHNIQUE

Abstract:
The olive (Olea europaea L.) is originated from Upper Mesopotamia which includes also South East Anatolia. There are many olive varieties whose morphologic and biologic characteristics are not known well. Some of them which had been identified by morphologic and biological characteristics, have some variations due to unknown genotypic or phenotypic causes. The growers choose the best variants, for top-working on their trees. In this research, trees of some standard Turkish cultivars (Ayvalik, Domat, Gemlik, Halhali, Kilis Yaglik, Manzanilla, Memecik, Nizip Yaglik and Tavsan Yuregi), which had been identified only by morphological characteristics, were selected (genetic polymorphism by RAPD (Random Amplified Polymorphic DNA)-PCR technique. 14 primers (Operon Technologies) were used and the results were confirmed by 3 replications. The diversity of RAPD patterns reflected genetic diversities among cultivars. However these data were also completed by some additional morphological and biological observations and analyses.
