PRODUCTION OF FERTILE INTERSPECIFIC SOMATIC HYBRID PLANTS BETWEEN SWEETPOTATO AND ITS WILD RELATIVE, IPOMOEA LACUNOSA

Abstract:
Embryogenic suspension culture protoplasts of sweetpotato cvs. Lizixiang, Tamayutaka and Xushu 18 were fused with petiole protoplasts of the wild relative Ipomoea lacunosa (K61) by polyethylene glycol (PEG) method. Fused protoplasts were cultured in a modified MS (Murashige and Skoog, 1962) medium containing 0.05 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5 mg/l kinetin. Ten to 12 weeks after plating, the obtained small calluses up to 1-2 mm in size were transferred to MS medium with 0.05 mg/l 2,4-D and 0.5 mg/l kinetin, resulting in rapid callus proliferation. The calluses were cultured on MS medium supplemented with 2 mg/l 6-benzylaminopurine (BAP) and further on the basal medium, they formed plants. Morphological, cytological, peroxidase isozyme and random amplified polymorphic DNA (RAPD) analyses showed that 108 regenerated plants were interspecific somatic hybrids of 3 cross-incompatible combinations. These somatic hybrids were fertile, and the hybrids between somatic hybrids of Lizixiang + I. lacunosa and Xushu 18 were also obtained.
