STUDIES ON THE POSSIBILITY OF PRODUCING VIRUS-FREE PLANTING MATERIAL OF PLUM CULTIVARS BY IN VITRO PROPAGATION

Abstract:
One of the major problems of plum growing is related to the viral disease Plum Pox virus (PPV). It is a limiting factor for spreading a number of plum cultivars with valuable economic and biological characteristics. On the other hand, the in vitro method enables the obtaining of great quantities of virus-free planting material for a shorter time, which is difficult to be achieved by the conventional methods.
The aim of the study was to work out the technological parameters for the production of PPV-free planting material of 4 plum cultivars on the basis of in vitro propagation and virology control.
The plum cultivars have been selected according to their economic significance and the relative resistance to PPV: cvs. ‘Cacanska lepotica’ and ‘Althan’s gage’, belonging to Prunus domestica L. species and cvs. ‘Santa Rosa’ and ‘Black Star’ of Prunus salicina L. species. The problems related to the major technological stages of the micropropagating process have been studied. The parameters of introducing the explants into sterile culture, the nutrient media and the conditions of multiplication and rooting and the adapting under in vivo conditions have been detected. In the process of studies a virology control of the source mother plants marked for explants, as well as of the obtained micropropagated and adapted plum plants has been conducted.
