OBSERVATIONS OF OVULES OF SOME PLUM CULTIVARS AFTER POLLINATION IN VITRO CONDITIONS

Abstract:
The protocol for pollination and fertilization followed by obtaining of the globular embryos in vitro conditions was elaborated during the many years’ investigations. For pollination in vitro it is important taking the non-pollinated flowers at the stage of ovule receptivity for pollen tube. Ovules should be pollinated on the solid White medium with addition 15% of sucrose. Twenty four hours after pollination the nucelluses isolated from pollinated ovules ought to be transferred onto Murashige and Skoog medium, Norstog medium, Beasley and Ting medium liquid medium with addition 3% of sucrose. Media for cultivation of fertilized nucelluses should be supplemented with growth substances – auxin IAA at concentration 1.0 mg ·l –1 and GA3 at concentration 0.2 – 0.5 mg ·l –1. The fertilized nucelluses should be cultivated in the test-tubes, in complete darkness, at temperature 25 o C, for 60 days.
