BIOLOGICAL AND IMMUNOENZYMIC CHARACTERISTICS OF PLUM POX POTYVIRUS ISOLATES ON STONE FRUITS IN BULGARIA

Abstract:
Plum Pox Virus (PPV) isolates, obtained from naturally infected Prunus sp. (peach, plum, apricot, cherry) trees, grown in collection- and commercial orchards of the Fruitgrowing Institute, Plovdiv and the Experimental Station, Silistra were characterized by biological (indicator) and immunoenzymic (DAS ELISA) tests.
The indicator peach (GF 305 and Elberta) and Myrobalan (GF 31) plants, inoculated by the method of chip budding under greenhouse conditions, induced typical PPV symptoms on leaves.
The results from mechanical inoculations of annual species, belonging to different botanical families, showed that PPV isolates infected 9 out of 11 species, tested at this stage - locally and systemically.
Serological tests were done with polyclonal antiserum (DAS ELISA) and two monoclonal strain specific antibodies, PPV-AL and PPV-4DG5 (DASI-ELISA). The monoclonal antibody - 5B, that recombined both PPV strains, was also used. The results obtained showed that the isolates belonged to PPV-M strain.
The immunological comparison of PPV isolates on stone fruits will be continued in our future work.
