THE EFFECT OF GROWTH RETARDANTS ON INDUCTION AND DEVELOPMENT OF GLORIOSA ROTHSCHILDIANA O'BRIEN TUBERS IN VITRO

Abstract:
Single shoots of Gloriosa rothschildiana O’Brien ‘Red Dark’ taken from aseptically grown shoot clusters were cultured at 220C under 16-hour photoperiod on the Murashige and Skoog (MS) medium supplemented with 6% of sucrose and different concentrations of flurprimidol (0.2, 1, 5 mg l-1) or paclobutrazol (0.01, 0.1, 1 mg l-1). The shoots were incubated for 12 weeks and then the number of tubers per explant, fresh weight and length of microtubers were recorded. Flurprimidol (0.2-1 mg l-1) and paclobutrazol (0.01-1 mg l-1) were found effective in inducing tuberization. The highest total fresh weight of tubers was achieved on the medium containing flurprimidol 0.2 mg l-1. Flurprimidol at concentration of 5 mg l-1 inhibited elongation of shoots and strongly reduced tuber formation. Growth retardants caused sprouting of tubers and flurprimidol at the level of 1 mg l-1 was the most efficient in promoting this process.
