SOMATIC EMBRYOGENESIS AND TRANSFORMATION IN NARCISSUS PSEUDONARCISSUS CULTIVARS

Abstract:
Somatic embryos (SEs) have been produced from bulb and shoot culture leaf explants of Narcissus pseudonarcissus cvs. Golden Harvest and St. Keverne. The highest yields of direct SEs were obtained from scape explants orientated with their basipetal surface away from the MS-based medium. SEs converted to plantlets most efficiently with a pre-treatment of 24 °C for 10-20 weeks on a medium with IBA, followed by transfer to 4 °C. These plantlets readily transferred to ex vitro conditions. Nodular callus was produced from scape explants of ‘Golden Harvest’ and ‘ St. Keverne’. This regenerated most successfully on a medium with TDZ in combination with either NAA or 2,4-D. Preliminary studies of the growth of callus in a liquid medium were promising. Transformation has been achieved via Agrobacterium tumefaciens, both with a wild type and an engineered strain.
