Abstract:
- In vitro Morphogenesis and Growth of Sinningia allagophylla (Gesneriaceae): Effects of Growth Regulators and Photoperiod
Explants obtained from nodal segments of seedlings of the brazilian cerrado plant Sinningia allagophylla grow in vitro were cultivated in Gamborg et al. 1968 medium (B5 medium) (half-strength nutrients) with pH 4.8 and supplemented with 3% sucrose (w/v) and plant growth regulators.
Three experiments were carried out in eight and eighteen hour photoperiod, using a factorial design: experiment 1 used combinations of benzyladenine (BA) (0.01 and 0.1 μg.ml-1) and kinetin (0.01 and 0.1 μg.ml-1) with indoleacetic acid (IAA) (0.1 μg.ml-1); experiment 2 used BA (0.01 and 0.1 μg.ml-1) with giberelic acid (GA3) (0.1 and 1.0 μg.ml-1) and experiment 3 used BA combinations (0.005 and 0.01 μg.ml-1) and of KIN (0.01 and 0.1μg.ml-1) with naphtaleneacetic acid (NAA) (0.01 and 0.1 μg.ml-1). The plant growth regulators BA and ANA at the lowest concentrations tested together or separately induced statistically significant increases in shoot and after root formation.
GA3 by itself acting only on shoot length growth.
GA3, on the other hand, hed altered BA morphogenetic effect: BA together with GA3 resulted in callus formation.
IAA and KIN did not present significant effects on the parameters measured.
General growth was promoted by short photoperiod (eight hour).
