ISOZYME CHARACTERIZATION OF GENETIC RESOURCES IN STRAWBERRY

Abstract:
Molecular markers are one of the best tools in genetic analysis for precise evaluation through traits that are expressed independently of environment. Seventy-four strawberry cultivars from diverse origins growing in our strawberry collection at CIFA Malaga were characterized through PAGE (Polyacrylamide gel electrophoresis) using polymorphic isozyme systems previously used in strawberry (PGI, LAP and PGM). The marker data were analyzed by simple matching and a dendrogram was based on the average linkage clustering (UPGMA). LAP zymograms (banding patterns) agreed well with previous work, PGI and PGM patterns are basically in agreement but some of them differed from those described for some cultivars and we have also found some new patterns. One of the LAP patterns is the most common (59%) in this collection. The combination of the three systems was not enough to achieve the complete identification of the whole collection but it was enough to establish the relationship between cultivars and it could be efficiently used as an additional criterion for verification of different cultivars. It has been possible to identity two large groups in the collection, corresponding to the geographic origins: North-Central-European versus Californian/Mediterranean.
