Abstract:
Full- and half-strength MS basal medium and full-strength BRP medium with various concentrations of auxin and cytokinin were tested on cotyledonary nodal explants of in vitro seedlings, in order to develop formulations to support growth, multiplication and rooting.
Rooting trials were also conducted with shoots from seedlings growing in vitro. The greatest number of shoots were induced on the cotyledonary explant in full-strength MS containing 2.5 mg/l BA. When shoots induced in the first stage were subcultured, shoot multiplication was best on half-strength MS whereas shoot elongation was greatest on hormone-free BRP. A maximum of 50% rooting was observed in hormone-free BRP medium following induction in 5.0 or 10.0 mg/l IAA for 10 days and in 0.5 and 5.0 mg/l IAA for 15 days.
