Abstract:
Shoot regeneration from leaf and stem explants of 19 Chrysanthemum cultivars (cut?, pot? and garden chrysanthemums) has been studied.
Adventitious shoots could be regenerated from leaf and stem explants of 16 cultivars.
Most cultivars showed the best shoot regeneration on Murashige & Skoog (1962) medium supplemented with 8.9 µM BAP (2.0 mg/l) and 2.7 µM NAA (0.5 mg/l). The number of regenerated shoots per explant was more than 10 in some of the cultivars studied.
It has not been posssible to regenerate shoots from leaf explants of cultivars 170 and 260. The cultivars 140 and 8010 produced the highest number of shoots.
Regenerated shoots can easily be rooted on half strength Murashige & Skoog medium supplemented with 0.98 µM IBA (0.2mg/l). After potting in soil and transfer to the glasshouse normal plants develop.
Apparently no somaclonal variation has been observed among the regenerants .
