IN VITRO ADVENTITIOUS SHOOT REGENERATION FROM VEGETATIVE APICES OF ALMOND AND OTHER PRUNUS SPECIES

Abstract:
A protocol for the induction of adventitious shoot formation from vegetative shoot apices of Prunus species was developed. Adventitious shoot regeneration was obtained with this method in almond, apricot, peach and plum. Apices, excised from in vitro shoots, were induced to regenerate for 30 days in darkness on an LP medium supplied with 8.8mM BA, 1.0mM NAA and 250mgl-1 cefotaxime, then transferred to a fresh auxin free medium, added with 1.4mM GA3, to the light and sub-cultured monthly. Histological studies performed on almond M55 revealed that the adventitious shoots originated from callus. The callus maintained shoot formation ability for months.
