Abstract:
Sugar beet shoot tips were cryopreserved with both an encapsulation-dehydration technique and a vitrification method.
With encapsulation-dehydration a three-step preculture and dehydration to 21 % water content proved to be optimal.
When applying vitrification loading of shoot tips at 20°C for 20 to 25 min and dehydration with PVS2 at 0°C for 20 to 30 min were optimal.
With both techniques, cold acclimation clearly improved survival. Finally, vitrification is recommended to cryopreserve sugar beet shoot tips since it proved to be easier and faster and surviving shoot tips resumed growth faster.
