AGROBACTERIUM-MEDIATED TRANSIENT EXPRESSION OF β-GLUCURONIDASE AND LUCIFERASE GENES IN LIMONIUM SINUATUM

Abstract:
We have developed an Agrobacterium-mediated transformation system for Limonium sinuatum. The shoot regeneration was observed on thidiazuron (TDZ)-containing medium. Shoot-forming capacity of this species depended on the cultivar. About 40 % of the explants of' cv. PF142 regenerated shoots on 0.1 µM TDZ medium. Luciferase (LUC) and alpha-glucuronidase (GUS) genes were introduced and transient expression of these gene were confirmed. Stable expression of the GUS gene was also confirmed, but shoot formation was strongly inhibited by hygromycin B, of which resistant gene was used as a selection marker.
