Abstract:
DNA analysis was performed on 18 Italian and foreign cultivars of Corylus avellana L. that are important either for table use or as processed kernels, and one cultivar of Corylus maxima Mill. with interesting characters.
Young leaf samples were obtained in a field collection located in Cravanzana (Piemonte, northwestern Italy), an important production region.
Duplicate samples of DNA were extracted from the leaves.
For the RAPD analysis, oligonucleotide primers (10-mers) from the kits A, I, J and M from Operon Technologies were tested.
All of the tested primers produced polymorphic bands between 500 and 3000 base pairs in size.
Based on a set of 45 polymorphic markers, the genotypes were compared and a phylogenetic tree was generated by cluster analysis.
The dendrogram had two major groups of cultivars.
In the second group were 'Tombul', 'Imperiale de Trebizonde', 'Jean's' and 'Fructo Rubro', all of which have tubular husks.
The other 15 cultivars were placed in the first group.
