The new nepovirus, previously isolated from reverted black currant, was inoculated back to healthy black currants.
Half a year after inoculation, the back-inoculated plants showed vein pattern symptoms very similar to the known acute symptoms of reversion disease, strongly suggesting that the virus was the causal agent of the disease.
On the basis of this result and the earlier results showing a close association between the virus and reversion disease, the virus was named Black currant reversion virus (BRV). Three and a half years after inoculation, in spring 2000, the first malformed flowers appeared in one back-inoculated black currant, thus proving that the isolated virus is the sole causal agent of reversion disease.
Different forms, or possibly strains, with symptoms varying from mild to very severe are known to occur in reversion disease.
As yet, the molecular determinants of the different disease phenotypes have not been established.
We have started the comparison of nine BRV isolates originating from six countries by comparing the 3’ non-translated region (1360 nt) nucleotide sequences of their RNA2 components.
Sequence analysis has indicated about 94-99% identity between the nine analysed isolates, with differences only in certain nucleotide positions.
