LONG TERM ASSESSMENT OF THE EFFECTS OF IN VITRO CHEMOTHERAPY AS A TOOL FOR APPLE STEM GROOVING VIRUS ELIMINATION

Abstract:
Shoot cultures of Malus domestica infected with apple stem grooving virus (ASGV) and treated in vitro with a combination of quercetin and ribavirin (10µg ml-1 each) for 9-12 weeks appear to have been successfully treated for the elimination of ASGV. The treated cultures were tested initially in March 1996, by immunocapture RT-PCR, and were found to be negative compared to the untreated controls which were all positive. The cultures testing negative were eventually subcloned to produce plants that were rooted and eventually planted in the field. Over three years after terminating treatment and with periodic monitoring by immunocapture RT-PCR, ASGV is still undetectable and the treated plants appear normal and slightly more vigorous than the untreated control plants. Monitoring for ASGV detection continues and the treated plants and fruit will be closely observed to determine if any phenotypic changes or abnormalities emerge.
